白細(xì)胞介素-1（interleukin-1,IL-1）是在感染和炎癥狀態(tài)下，由多種細(xì)胞產(chǎn)生的、有多方面生物學(xué)功能的細(xì)胞因子，其生物學(xué)作用十分廣泛，主要包括參與介導(dǎo)炎癥反應(yīng)、調(diào)節(jié)免疫、調(diào)節(jié)機(jī)體代謝。IL-1分子重量為17.5KD，有兩種類型：IL-1α和IL-1β，主要以IL-1β為主。IL-1受體（interleukin-1 receptor，IL-1R）包括Ⅰ型和Ⅱ型，此外還存在一種內(nèi)源性IL-1受體拮抗劑 （interleukin-1 receptor antagonist，IL-1ra或IL-1γ），IL-1ra可以與IL-1受體結(jié)合，但是不引起相應(yīng)的作用。炎癥情況下,粒細(xì)胞巨噬細(xì)胞集落刺激因子(GM-CSF) 水平升高，它也能促進(jìn)IL-1 的上調(diào)作用,增加IL-1β的合成;另外關(guān)節(jié)滑膜細(xì)胞也可自分泌IL-1β。
人白介素-1β重組蛋白是在大腸桿菌中重組表達(dá)的，是一條非糖基化的多肽鏈，包含有153個(gè)氨基酸，分子量為17000Da。該人白介素-1β重組蛋白最終由標(biāo)準(zhǔn)的色譜技術(shù)純化出。

溶解：提供的人白介素-1β (IL 1 beta)產(chǎn)品為過(guò)濾滅菌的白色凍干粉末；該凍干粉末是從含1mg/ml人白介素-1β (IL 1 beta)的50mM 磷酸緩沖液（pH 7.1）和150mM NaCl溶液中過(guò)濾滅菌并凍干產(chǎn)生的。我們建議加無(wú)菌的18MΩ-cm水以配制工作儲(chǔ)存液到100ug/ml，后繼實(shí)驗(yàn)過(guò)程中可根據(jù)實(shí)驗(yàn)要求進(jìn)一步稀釋。

保存：人白介素-1β (IL 1 beta)凍干粉末在室溫可保存3周，或置于-18℃凍存用于長(zhǎng)期保存。重新溶解后可以在4℃冷藏保存至少2-7天；如果想延長(zhǎng)保存時(shí)間，建議分裝（加載體蛋白如0.1% HSA或 BSA）后置于-18℃凍存。請(qǐng)避免反復(fù)凍融。

Introduction 

Interleukin-1b is produced by activated macrophages, IL-1B stimulates thymocyte proliferation by inducing il-2 release, b-cell maturation and proliferation, and fibroblast growth factor activity. IL1B proteins are involved in the inflammatory response, being identified as endogenous pyrogens, and are reported to stimulate the release of prostaglandin and collagenase from synovial cells.

Description 

Interleukin-1 beta Human Recombinant produced in E.Coli is a non-glycosylated, Polypeptide chain containing 153 amino acids and having a molecular mass of 17000 Dalton.
The IL-1b is purified by proprietary chromatographic techniques.

Physical Appearance 

Sterile Filtered White lyophilized (freeze-dried) powder.

Formulation 

The protein was lyophilized from a concentrated (1mg/ml) sterile solution containing 50mM Phosphate buffer pH=7.1 and 150mM NaCl.

Solubility 

It is recommended to reconstitute the lyophilized Interleukin-1b in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability  （穩(wěn)定性）

Lyophilized Interleukin-1b although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution IL1b should be stored at 4°C between 2-7 days and for future use below -18°C.For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please prevent freeze-thaw cycles.

Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by SDS-PAGE.

Amino acid sequence （氨基酸序列）

The sequence of the first five N-terminal amino acids was determined and was found to be Ala-Pro-Val-Arg-Ser.

Biological Activity  （生物活性）

The specific activity as determined in the test of augmentation of lymphocyte proliferation assay using mouse thymus was found to be 200,000,000 IU/mg.

Protein content  （蛋白質(zhì)含量）

Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 0.631 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a calibrated solution of IL-1 beta Recombinant as a Reference Standard.
