白細(xì)胞介素-2(IL-2)是一種最早發(fā)現(xiàn)的具有廣泛生物活性的細(xì)胞因子，它必須和IL-2受體結(jié)合才能發(fā)揮生物學(xué)效應(yīng)。白細(xì)胞介素-2受體(IL-2R)是由3個(gè)糖蛋白亞基組成的，分別為 α，β，γ鏈，分子量為55kD，75kD和64kD，故又稱p55，p75和p64。單個(gè)亞基不能傳遞IL-2信號(hào)，只有2或3個(gè)亞基組成復(fù)合體方能發(fā)揮受體功能。大量交叉實(shí)驗(yàn)表明，IL―2的種屬特異性是有一定規(guī)律的，一般呈下行性，即進(jìn)化程度高的動(dòng)物的IL-2可以作用于進(jìn)化程度較低的動(dòng)物。如人和猿的IL-2。幾乎可以作用于所有哺乳動(dòng)物的細(xì)胞；但其它哺乳動(dòng)物的IL-2很少可以作用于人。由于IL-2的生物學(xué)作用是以刺激T細(xì)胞增生為主，故又稱T細(xì)胞生長(zhǎng)因子，除此之外還有其它重要功能。
人白介素-2重組蛋白是在大腸桿菌中重組表達(dá)的，是一條單鏈、非糖基化的突變蛋白，包含有134個(gè)氨基酸，其中在126位的Cys氨基酸被替換成了Ser，分子量為15517Da。該人白介素-2重組蛋白最終由標(biāo)準(zhǔn)的色譜技術(shù)純化出。

溶解：提供的人白介素-2(IL 2)產(chǎn)品為過(guò)濾滅菌的白色凍干粉末；該凍干粉末是從含1.1mg/ml人白介素-2(IL 2)的 0.17mg磷酸二氫鈉和0.89mg磷酸氫二鈉緩沖液（pH 7.5）中經(jīng)進(jìn)一步透析并凍干產(chǎn)生的。我們建議加無(wú)菌的20mM AcOH以配制工作儲(chǔ)存液到100ug/ml，后繼實(shí)驗(yàn)過(guò)程中可根據(jù)實(shí)驗(yàn)要求進(jìn)一步稀釋。
保存：人白介素-2(IL 2)凍干粉末在室溫可保存3周，或置于-18℃凍存用于長(zhǎng)期保存。重新溶解后可以在4℃冷藏保存至少2-7天；如果想延長(zhǎng)保存時(shí)間，建議分裝（加載體蛋白如0.1% HSA或 BSA）后置于-18℃凍存。請(qǐng)避免反復(fù)凍融。

Introduction 

IL2 is a secreted cytokine that is important for the proliferation of T and B lymphocytes. The receptor of this cytokine is a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL4) and interleukin 7 (IL7). The expression of this gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a similar gene in mice leads to ulcerative colitis-like disease, which suggests an essential role of this gene in the immune response to antigenic stimuli.

Description 

Interleukin-2 Human Recombinant produced in E.Coli is a single, non-glycosylated mutein (variant form) of human IL-2 polypeptide chain containing 134 amino acids and having a molecular mass of 15517 Dalton.
Our Interleukin -2 has an Ser substitute for Cysteine at position 126.
The IL-2 is purified by proprietary chromatographic techniques.

Physical Appearance 

Sterile Filtered White lyophilized (freeze-dried) powder.

Formulation 

The protein (1.1mg/ml) was lyophilized after extensive dialysis against 0.17mg sodium monobasic & 0.89mg dibasic sodium phosphate buffer to a pH=7.5.

Solubility 

It is recommended to reconstitute the lyophilized Interleukin 2 in sterile 20mM AcOH not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability  （穩(wěn)定性）

Lyophilized Interleukin-2 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution IL2 should be stored at 4°C between 2-7 days and for future use below -18°C.
For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please prevent freeze-thaw cycles.

Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by SDS-PAGE.

Amino acid sequence   （氨基酸序列）

The sequence of the first five N-terminal amino acids was determined and was found to be Met-Ala-Pro-Thr-Ser.
Biological Activity  The ED50 as determined by the dose-dependant stimulation of murine CTLL-2 cells is < 0.0645 ng/ml, corresponding to a Specific Activity of 15,500,000IU/mg.
Protein content  Protein quantitation was carried out by two independent methods
1. UV spectroscopy at 280 nm using the absorbency value of 0.614 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).

2. Analysis by RP-HPLC, using a calibrated solution of IL-2 as a Reference Standard.
