The Complete Kit for Human CD4+CD25+ Regulatory T Cells is a simple two-step method that is designed to isolate CD4+CD25+ cells from whole blood or buffy coat. CD4+ T cells are enriched from whole blood or buffy coat by negative selection using RosetteSep® CD4+ T Cell Enrichment Cocktail. The RosetteSep® cocktail contains a combination of antibodies that is directed against non-CD4+ T cells and glycophorin A red blood cells (RBCs). When centrifuged over a buoyant density medium such as RosetteSep® DM-L (Catalog #15705) or Ficoll-Paque™ PLUS (Catalog #07957), the unwanted cells pellet along with the RBCs. The purified cells are present as a highly enriched population at the interface between the plasma and the buoyant density medium. The enriched CD4+ T cells are then selected for CD25+ cells by positive selection using EasySep® Human CD25 Positive Selection Kit. Cells are targeted with Tetrameric Antibody Complexes recognizing CD25 and dextran-coated magnetic particles. Labeled cells are separated using an EasySep® magnet without the use of columns. Cells of interest remain in the tube while unwanted cells are poured off.
