Stemcell 19757 EasySep™ Mouse Mammary Stem Cell Enrichment Kit旨在從新鮮解離的小鼠乳腺組織中分離出乳腺干細(xì)胞。不需要的細(xì)胞被針對(duì)非上皮細(xì)胞(CD45, CD31, TER119, BP-1)的生物素化的抗體瞄準(zhǔn)去除。標(biāo)記的細(xì)胞然后被確認(rèn)針對(duì)抗生物素和葡聚糖的四聚體的抗體配合物識(shí)別。這些細(xì)胞被綁定到磁性粒子并使用沒有柱的EasySep™磁鐵分離。未受破壞的上皮細(xì)胞富集餾分傾到一個(gè)新的管中，然后用CD24和CD49f抗體標(biāo)記，這樣，CD24+CD49f++ 子種群就可以通過熒光激活細(xì)胞分選分離。

FACS Profile Results with EasySep™ Mouse Mammary Stem Cell Enrichment Kit

Freshly dissociated mouse mammary epithelial cells are distributed according to their expression of CD49f and CD24. The mammary repopulating unit (MRU)-rich fraction is found in the CD49f++CD24+ subset. The mammary colony-forming cells (Ma-CFCs) are found in the CD49f+ CD24++ subset. In certain strains of mice (such as C57BI/6J) there can be a small number of CD24++CD49f++ cells (indicated * in the dot plot). Although mammary stem cells have been detected in this fraction, their frequency is much lower than in the MRU fraction.
