紅細(xì)胞生成素（erythropoietin，EPO）對(duì)紅細(xì)胞生成的特異性刺激作用的細(xì)胞因子。紅細(xì)胞生成素是一種糖蛋白，在胎兒體內(nèi)由腎臟及肝臟產(chǎn)生，而在成人體內(nèi)主要由腎臟產(chǎn)生。目前已知人紅細(xì)胞生成素有兩種存在形式，即人EPO-α及人EPO-β，二者氨基酸組成及順序相同，都含有165個(gè)氨基酸殘基。分子量、等電點(diǎn)及生物活性也都類似，差別在于二者的糖型組成不同，EPO-α含有較多的N-乙酰氨基葡萄糖和N-乙酰神經(jīng)氨酸，總的含糖量也較EPO-β高。紅細(xì)胞生成素-α是調(diào)節(jié)紅細(xì)胞增殖、成熟和分化的一種造血生長因子。

 人紅細(xì)胞生成素-α重組蛋白是在CHO中國倉鼠卵巢細(xì)胞中重組表達(dá)的，是一條單鏈的多肽鏈，包含有165個(gè)氨基酸，計(jì)算分子量為21，000Da，由于發(fā)生糖基化修飾而導(dǎo)致實(shí)際分子量為30，400 Da。該人紅細(xì)胞生成素-α重組蛋白最終由標(biāo)準(zhǔn)的色譜技術(shù)純化出。

溶解：提供的人紅細(xì)胞生成素-α(EPO a)產(chǎn)品為過濾滅菌的白色凍干粉末；該凍干粉末中 人紅細(xì)胞生成素-α、檸檬酸鈉、NaCl、檸檬酸的比率分別為：1mg、0.58mg、0.58mg、0.006 mg。我們建議加無菌的18MΩ-cm水以配制工作儲(chǔ)存液到100ug/ml，后繼實(shí)驗(yàn)過程中可根據(jù)實(shí)驗(yàn)要求進(jìn)一步稀釋。

保存：人紅細(xì)胞生成素-α(EPO a)凍干粉末在室溫可保存3周，或置于-18℃凍存用于長期保存。重新溶解后可以在4℃冷藏保存至少2-7天以上；如果想延長保存時(shí)間，建議分裝（加載體蛋白如0.1% HSA或 BSA）后置于-18℃凍存。請(qǐng)避免反復(fù)凍融。

Introduction 

This gene is a member of the EPO/TPO family and encodes a secreted, glycosylated cytokine composed of four alpha helical bundles. The protein is found in the plasma and regulates red cell production by promoting erythroid differentiation and initiating hemoglobin synthesis. This protein also has neuroprotective activity against a variety of potential brain injuries and antiapoptotic functions in several tissue types.

Description 

Erythropoietin-alpha Human Recombinant is produced in Chinese hamster ovary (CHO) cells by recombinant DNA technology is a single, polypeptide chain containing 166 amino acids and having a predicted molecular mass of 21,000 Dalton and apparent glycosylated molecular mass of 36-40kDa. EPO-a is purified by proprietary chromatographic techniques.

Physical Appearance 

Sterile Filtered White lyophilized (freeze-dried) powder.

Formulation 

Each mg of lyophilized powder contains 0.59 mg sodium citrate, 0.58 mg sodium chloride and 0.006 mg citric acid.

Solubility 

It is recommended to reconstitute the lyophilized Epoetin-a in sterile 18M-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability 

Lyophilized Erythropoietin-a although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution EPO-alpha should be stored at 4°C between 2-7 days and for future use below -18°C.For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).

Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by SDS-PAGE.

The Specific Activity was measured by Normocyth -aemic mice and was found to be 150,000 IU/mg.
