GDNF promotes the survival and differentiation of dopaminergic neurons in culture，and is able to prevent apoptosis of motor neurons induced by axotomy. The encoded protein is processed to a mature secreted form that exists as a homodimer. The mature form of the protein is a ligand for the product of the RET (rearranged during transfection) protooncogene. In addition to the transcript encoding GDNF，two additional alternative transcripts encoding distinct proteins，referred to as astrocyte-derived trophic factors，have also been described. Mutations in this gene may be associated with Hirschsprung disease.
GDNF enhances survival and morphological differentiation of dopaminergic neurons and increases their high-affinity dopamine uptake.

溶解： 提供的人神經(jīng)膠質(zhì)源神經(jīng)營(yíng)養(yǎng)因子 (GDNF)產(chǎn)品為過(guò)濾滅菌的白色凍干粉末；該凍干粉末是從含人神經(jīng)膠質(zhì)源神經(jīng)營(yíng)養(yǎng)因子 (GDNF)的10mM檸檬酸鈉和150mM NaCl溶液中經(jīng)透析并凍干產(chǎn)生的。我們建議加無(wú)菌的18MΩ-cm水以配制工作儲(chǔ)存液到100ug/ml，后繼實(shí)驗(yàn)過(guò)程中可根據(jù)實(shí)驗(yàn)要求進(jìn)一步稀釋。
保存：人神經(jīng)膠質(zhì)源神經(jīng)營(yíng)養(yǎng)因子 (GDNF)凍干粉末在室溫可保存3周，或置于-18℃凍存用于長(zhǎng)期保存。重新溶解后可以在4℃冷藏保存至少2-7天以上；如果想延長(zhǎng)保存時(shí)間，建議分裝（加載體蛋白如0.1% HSA或 BSA）后置于-18℃凍存。請(qǐng)避免反復(fù)凍融。

Glial derived Neurotrophic Factor Human Recombinant produced in E.Coli is a homodimer, non-glycosylated, polypeptide chain containing 2 x 135 amino acids and having a total molecular mass of 30,360 Dalton.
GDNF is purified by proprietary chromatographic techniques.

Sterile Filtered White lyophilized (freeze-dried) powder.

Formulation 

GDNF was lyophilized after dialysis against 10mM sodium citrate and 150mM NaCl.

Solubility   （溶解性）

It is recommended to reconstitute the lyophilized Glial Derived Neurotrophic Factor in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability  （穩(wěn)定性）

Lyophilized Glial-derived Neurotrophic Factor although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution GDNF should be stored at 4°C between 2-7 days and for future use below -18°C.
For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please prevent freeze-thaw cycles.

The sequence of the first five N-terminal amino acids was determined and was found to be Met-Ser-Pro-Asp-Lys.

Biological Activity  （生物活性）

The ED50, calculated by the dose-dependant dopamin uptake in rat mesencephalic cultures was found to be 5-10 ng/ml, corresponding to a specific activity of 100,000-200,000Units/mg.
