激活素(activins) Activin A是轉(zhuǎn)化生長因子(TGF)β家族的一種分泌蛋白，可控制胚胎軸發(fā)育為有功能的前腸源性組織，調(diào)解多種細(xì)胞的生長和分化。多種研究表明，其能誘導(dǎo)胰腺β細(xì)胞的分化。尤其與其他因子的聯(lián)合作用后能發(fā)揮更顯著的誘導(dǎo)作用。如activin A聯(lián)合全反式維甲酸可以更有效地誘導(dǎo)鼠ESC為產(chǎn)胰島素細(xì)胞，而activin與BTC共同作用可以轉(zhuǎn)化新生大鼠導(dǎo)管細(xì)胞為產(chǎn)胰島素細(xì)胞。卵泡抑素(follistatin)和表皮生長因子相關(guān)肽(Cripto)是ac2tivins的天然拮抗劑，有研究表明了胰島分化階段導(dǎo)管上皮細(xì)胞里的activins及其抑制因子follistatin和Cripto都有所增加,卵泡抑素抑制activin的同時(shí)，能顯著促進(jìn)胰腺導(dǎo)管細(xì)胞新生而減少β細(xì)胞的分化，故選擇抑制activin信號(hào)途徑可運(yùn)用于擴(kuò)增前體細(xì)胞，而激活activin有利于β細(xì)胞的分化。這些結(jié)果表說明,胰腺生長和分化之間的平衡可通過activin信號(hào)途徑的精確調(diào)節(jié)來實(shí)現(xiàn)。同時(shí)它還被廣泛用于干細(xì)胞研究，用來誘導(dǎo)分化的胚胎干細(xì)胞形成內(nèi)胚層。

Activin A is a member of the TGFβ superfamily. It is a 26 kDa disulfidelinked homodimer of two βA chains, each containing 116 amino acid residues. Activin A is widely used in stem cell research. It has been used to induce the differentiation of embryonic stem cells to the definitive endoderm. The biological activities of Activin A can be neutralized by diffusible TGF-β antagonists, Follistatin (also available from StemRD). StemRD’s Activin A is made in human 293 cells, which renders human-specific post-translational modifications. We purified this product through a series of chromatography including HPLC to near homogeneity.

? Biological Activity: The activity was determined by using a SMAD reporter gene assay in human 293 cells. The EC50 ranges from 0.5 – 2.0 ng/ml
